CHECK-LIST Criteria of statement assessment for definition of poliomyelitis virus type

¹ Criteria of Assessment
State the purpose of the color test for definition of poliomyelitis virus type. Completely describe the steps of the color test and name the components of the reaction
Add 0,25 ml of a virus content material to each of 3 test tubes with culture cells and label I, II, III
Add 0,25 ml of poliomyelitis type I, II, or III antiserum in the corresponding test tubes
Add 0,6 ml vaseline oil to each tube. Incubate for 48-72 hours.
Comment on the results. Explain the reaction mechanism.

CHECK-LIST Criteria of statement assessment of direct immunofluorescence assay (DIFA)

¹ Criteria of Assessment
State the purpose of the DIFA. Completely describe the steps of the DIFA and name the components of the reaction
Make preparation containing unknown test microorganisms (Ag) on a special microscope slide with shallow wells. Make positive and negative control wells also
Fix it by liquid clamp (acetone).
Add fluorescent-labeled immunoglobulins specific to Ag tested. Incubate 30 min
Remove unbound immunoglobulins by washing solution
Dry slide and view with a microscope using ultraviolet lamp (fluorescent microscope). With a positive reaction the target antigen fluorescence is observed. Compare test wells with Positive control. Negative control well remains dark
Comment on the received results. Explain the reaction mechanism

CHECK-LIST Criteria of statement assessment of polymerase chain reaction (PCR)

¹ Criteria of Assessment
State the purpose of PCR. Completely describe the steps of the PCR and name the components of the reaction
Test DNA is heated to 95oC and complementary strands of DNA are separated. This stage is called DNA denaturation.
Target-specific oligonucleotide primers bind to each strand. This stage is called as "annealing" (accession).
Thermostable DNA polymerase completes complementary chains of DNA. This stage is called "elongation" (lengthening).
The three stages are repeated 30-40 times (cycles) that amplify each original gene fragment to over 1 million copies
Explain the reaction mechanism

CHECK-LIST Criteria of statement assessment of reactions of the enzyme immunoassay (EIA) (IFA in Russian)

¹ Criteria of Assessment
State the purpose of EIA. Completely describe the steps of the EIA and name the components of the reaction
Specific antigen is adsorbed in wells of a microtiter plate. Add test serum and incubate 30 minutes. Include positive and negative wells also
Remove unbound immunoglobulins by washing solution
Add hydrogen peroxidase-labeled anti-human immunoglobulins. Incubate 30 minutes
Add liquid substrate and keep in dark for 10-15 minutes. The substratum (hydrogen peroxide) reacts with the indicator (a diamine benzidine) to cause a color change whose intensity is measurable with spectrophotometer
Comment on the result: principle of estimation of intensity of coloring. Explain the reaction mechanism
