Remove unbound immunoglobulins by washing solution
Dry slide and view with a microscope using ultraviolet lamp (fluorescent microscope). With a positive reaction the target antigen fluorescence is observed. Compare test wells with Positive control. Negative control well remains dark

¹ Criteria of Assessment
State the purpose of PCR. Completely describe the steps of the PCR and name the components of the reaction
Test DNA is heated to 95oC and complementary strands of DNA are separated. This stage is called DNA denaturation.
Target-specific oligonucleotide primers bind to each strand. This stage is called as "annealing" (accession).
Thermostable DNA polymerase completes complementary chains of DNA. This stage is called "elongation" (lengthening).
The three stages are repeated 30-40 times (cycles) that amplify each original gene fragment to over 1 million copies

Add liquid substrate and keep in dark for 10-15 minutes. The substratum (hydrogen peroxide) reacts with the indicator (a diamine benzidine) to cause a color change whose intensity is measurable with spectrophotometer
